This article, though short, is already in direct disagreement with DNA_polymerase_I, which says that the 3=>5 proofreading activity is removed in the Klenow fragment. I will look this up and try to resolve it. Capybara 19:44, 14 November 2005 (UTC)Reply


The NEB book (new england biolabs enzyme catalog) says the following: Klenow fragment is a proteolytic product of E.colie DNA Polymerase I which retaing polymerization and 3' -> 5' exonuclease activity , but has lost 5' -> 3' exonuclease activity. Klenow retains the polymerization fidelity of the holoenzyme without degrading 5' termini.

Yeah that's right the Klenow fragment has no 5' to 3' exonuclease activity. It's used in the labelling method called 'random primer method' --User:Matt.S 20:00, 30 December 2005 (UTC)Reply

I decided this article was ridiculously poor, being both technically incorrect and lacking any detail on how you make a Klenow fragment and why you would want to. Felt I had to do something about it so wrote a bit of an article that's technically correct and has a bit more information. Though the wordings not the best in places and the formatting is probably shoddy, as i don't now how to do it. Also i would have added a few pictures but can't cos i'm not a user and wasn't sure about the copyrighting and that.
