Most of the county routes in Erie County, New York, act as primary roads in the less developed areas and also serve to interconnect the various villages and hamlets of the county. Municipalities served include the cities of Buffalo and Tonawanda, and the villages of Angola, Evans, Farnham, Lancaster, Springville, East Aurora and Depew. Routes 545 to 580 are listed below.

County Route 546 is a north–south route within the town of Brant. Route 546 begins at its south end at the Collins town Line. Route 546 heads north as Mile Block Road through the town of Brant intersecting Mile Strip Road (CR 93), then North Collins Road (NY 249) before coming to its north end at Cain Road (CR 491) which is actually on the EvansBrant town line.

Mile Block Road actually begins at Seneca Road in the town of Collins and within the Cattaraugus Indian Reservation. Since generally there are no county routes within Indian Reservations, Route 546 begins once Mile Block Road leaves the reservation, and at the same time leaves Collins into the town of Brant.

County Route 547, named Two Rod Road, is a north–south route in the towns of Alden and Marilla. Route 578 begins at its southern end at an intersection with Clinton Street (NY 354) and Two Rod Road (unsigned NY 954G and former NY 358) in Marilla. Two Rod Road continues south from here as NY 954G. Route 547 travels north into the town of Alden, crossing Cayuga Creek and then intersecting Broadway (US 20) within the town. Route 547 then comes to an end at Westwood Road (CR 343).

Route 548 is 3.73 miles (6.00 km) long and travels from West River Parkway (unsigned NY 957C) at its west end to East River Road (CR 11) at its east end. Route 548 intersects well traveled Baseline Road (CR 249) at 2.67 miles (4.30 km) and well-traveled South Parkway (unsigned NY 957B) at 3.17 miles (5.10 km).

County Route 550 is an east–west route named Kenmore Avenue in the city of Buffalo from its west end at the Tonawanda town line to its east end at Military Road (NY 265).

Kenmore Avenue is numbered as three separate county routes along its path: CR 189 in the town of Tonawanda from Grand Island Boulevard (unsigned NY 950C) to the Buffalo city line, CR 550, and CR 307 from NY 265 to Main Street (NY 5) at the AmherstBuffalo line.

County Route 551, named Darien Road, is an east–west route within the town of Colden. Route 551 begins at its west end at Boies Road (CR 541), intersects Center Street (CR 33) at 0.2 miles (0.32 km) and then travels east to its ends at Lewis Road (CR 380).

Darien Road itself actually is 4.2 miles (6.8 km) long and starts at Hayes Hollow Road (CR 397) in Colden; and actually ends at Olean Road (NY 16) at the WalesHolland town line; Route 551 is only a portion of this named road.

County Route 552 is a north–south route in the towns of Boston and Orchard Park. Named Chestnut Ridge Road, Route 552 begins at Boston State Road (CR 227) at its southern end in Boston. Route 552 then travels to its north end just inside the town of Orchard Park at the intersection of Boston Ridge and Chestnut Ridge roads. NY 277 enters this junction from the southwest on Boston State Road and leaves to the north as Chestnut Ridge Road.

County Route 553, named Transit Road Extension, is a north–south route in the town of Orchard Park. Route 553 begins at its south end at Jewett–Holmwood Road (CR 368). Transit Road then heads north to its end at an intersection with Quaker Road (US 20A). Transit Road continues as NY 187 north from here. Route 553 runs along the border between the towns of Orchard Park and Aurora.

County Route 554, named Youngs Road, begins at Aero Drive (CR 324) near the southwest–northeast runway for Buffalo-Niagara International Airport in the town of Cheektowaga. Just after beginning, on the north side of the New York State Thruway overpass, Route 554 enters the town of Amherst; however, the area is known as Williamsville because of its proximity to the village. Youngs Road intersects Wehrle Drive (CR 290), a busy east–west route in Amherst at an 8th of mile. After that intersection, CR 554 heads north and is bordered on its east side by Erie Community College (North Campus) all the way to its north end at Main Street (NY 5).

County Route 555 is a short east–west route in the town of Sardinia named Dutchtown Road and is only two-tenths of a mile long. Route 555 begins at Olean Road (NY 16) and ends at South Protection Road (CR 531).

County Route 557, named Mill Street, is a north–south route in the town of Amherst. Route 557 begins at its southern end at the Williamsville village line and ends 0.6 miles (0.97 km) later at NY 324 (Sheridan Drive). Mill Street itself begins at Main Street (NY 5) in the village of Williamsville.

County Route 559

County Route 559
Location: Clarence
Length: 5.8 mi (9.3 km)

County Route 559 is the portion of Tonawanda Creek Road that lies in Clarence. Route 559 runs east–west beginning at its west end at its intersection with Transit Road (NY 78) in an area of Clarence called Millersport. Tonawanda Creek Road continues west of NY 78 as CR 2. Route 559 travels east to its end at Rapids Road (CR 43) and Salt Road (CR 560). CR 559 was once part of NY 268, which continued south onto Salt Road.[citation needed]

In 2008, work was started on replacing a section of Tonawanda Creek Road in Clarence.[2] This is because of detours for travelers when they intersect Transit Road. There was a cave-in on CR 559 in June 2004[2] and has since been repaired as of mid-2009.[citation needed]

County Route 560, named Salt Road, is a major thoroughfare through and north of Clarence. The route is 6.67 miles (10.73 kilometres) long, stretching from NY 5 in Clarence to CR 43 and 559 north of Clarence Center. The route is part of the old alignment of NY 268, which included the adjacent CR 559.

County Route 562 is an east–west route within the town of Colden. Named Heath Road, Route 562 begins at the intersection of State Road (NY 240) and Boston–Colden Road (CR 222) at its west end in the hamlet of Colden within the town of Colden. Route 562 then travels east through Blanchard Hill to its east end at Hayes Hollow Road (CR 397). Heath Road continues east past the intersection with Hayes Hollow another 1.4 miles (2.3 km) to Center Street (CR 33).

County Route 563 is an east–west route that begins in the town of Colden and ends in the town of Sardinia. Route 563, named Foote Road, begins at the intersection of Glenwood – East Concord Road (NY 240 and CR 30) and Morse Road (CR 433). Route 563 comes to its east end in Sardinia at Pratham Road (CR 407).

County Route 571

County Route 571
Location: West Seneca
Length: 3.7 mi[1] (6.0 km)

County Route 571 is the designation for a county-maintained section of NY 16 in the town of West Seneca. The route follows Center Road, an east–west road that begins and ends at Seneca Street (CR 215) at points about 4 miles (6.4 km) apart. The road runs alongside the Aurora Expressway (NY 400) and loosely parallels Seneca Street as it heads east. This section of NY 16 intersects the busy Union Road (NY 277). CR 571 also connects to Lein (CR 154) and Bullis roads at its eastern terminus, located in an area of town called East Seneca. NY 16 continues onto Seneca Street at both termini.

County Route 572

Length: 5.5 mi[1] (8.9 km)

County Route 572 is the designation for a county-maintained section of NY 16 and NY 78 in the towns of Aurora and Elma and the village of East Aurora. The route begins where NY 16 and NY 78 split from US 20A in East Aurora and it travels north as Buffalo Street in East Aurora, Buffalo Road in Aurora, and Seneca Street in Elma to Transit Road (US 20). This section of NY 16 and NY 78 connects to several local roads, including Jamison Road (CR 574 and former NY 422).

At CR 572's north end, NY 78 turns north onto Transit Road to follow US 20 while NY 16 continues west on Seneca Street into West Seneca. This segment of NY 16 is also county-maintained as CR 215.

County Route 573 is an east–west route in the village of Akron and town of Newstead. Route 573 begins at its western end at Cedar Street (NY 93) in Akron and is named John Street. Route 573 travels east and turns northeast onto Bloomingdale Road and travels into the town of Newstead. Route 573 then comes to an end at the Genesee County Line. CR 573 was once the western half of NY 267, which ran from Akron to Basom (a hamlet in Alabama). The NY 267 designation was eliminated on August 14, 1980, and replaced by CR 573 in Erie County.

County Route 574 is a major thoroughfare north of East Aurora. Known locally as Jamison Road, the road supplies the hamlet of East Elma. The road starts at Seneca Street (NY 16, NY 78, and CR 572) south of the hamlet Spring Brook and ends at Two Rod Road (unsigned NY 954G) in the town of Marilla. This county route was formerly NY 422, but this designation was eliminated on September 25, 1980, and the road was given to the county.

County Route 575 is an east–west route (northwest–southeast) in the town of Hamburg named Bayview Road. Route 575 begins at its east end at South Park Avenue (US 62). Route 575 then travels a short distance west to its end at Big Tree Road (CR 576). Bayview Road continues northwest past Big Tree Road (CR 576) and the section of Bayview Road located east of McKinley Parkway (CR 204) in Hamburg is numbered CR 169.

County Route 576, named Big Tree Road, is an east–west route in the town of Hamburg. Route 576 begins at its west end at Saint Francis Drive (NY 75) near NY 5 in an area of town called Athol Springs.[3] Route 576 known as Big Tree Road, heads east intersecting only local roadways until in comes to its east end at Bayview Road (CR 575).

Big Tree Road continues east past Bayview Road (CR 575) as just Bayview Road and begins as US 20A at Southwestern Boulevard (US 20) in Hamburg.

County Route 580, named Ellicott Creek Road, is an east–west route entirely in the town of Tonawanda. Route 580 begins at the Tonawanda city line and travels to Niagara Falls Boulevard (US 62) its east end. Niagara Falls Boulevard divides the town of Tonawanda and town of Amherst. There is an intersection with Colvin Boulevard (CR 303) within the town. Ellicott Creek Road continues west into the city of Tonawanda and ends at Young Street.
